David Smyth was awarded a BSc (Adelaide) in 1966, and a PhD from ANU in 1970, and worked as a Research Scientist at the Medical Research Council Cytogenetics Unit in Edinburgh before being appointed to Lectureship in the Monash department of Genetics in 1974. He was promoted to Senior Lecturer in 1980 and to Reader in 1992, and in November 2000 was appointed to a Personal Chair in Genetics. The title of Emeritus was conferred on Professor Smyth following his retirement in 2010.
